چکیده

BACKGROUND Indole-3-carbinol (I3C) is a naturally occurring phytochemical found in cruciferous vegetables. The aim of the present study is to investigate the influence of I3C on radiosensitivity in epidermal growth factor receptor (EGFR)-positive and EGFR-negative lung cancer cell lines. METHODS Human lung adenocarcinoma NIH-H1975 cells and human lung squamous carcinoma NIH-H226 and NIH-H520 cells were routinely cultured in RPMI-1640. MTT assay and clonogenic assay were used to detect cell growth and survival, respectively. Western blot and RT-PRC assay was employed to detect EGFR protein and mRNA expression. RESULTS 5 μmol/L of I3C significantly reduced radiosensitivity of EGFR-positive NIH-H1975 and NIH-H226 cells, but failed to affect radiosensitivity of EGFR-negative NIH-H520 cells. Furthermore, I3C caused an increased expression of total EGFR and pEGFR (Y845) protein in NIH-H1975 and NIH-H226 cell lines, but not in NIH-H520 cell line. A reduction of EGFR expression by EGFR-siRNA significantly inhibited I3C-caused radioresistance in NIH-H1975 cells. CONCLUSIONS Our data presented here for the first time demonstrate that I3C reduces radiosensitivity of lung cancer cells by mediating EGFR expression, indicating that EGFR may be an important target for I3C-mediated radioresistance in lung cancer.

The role and mechanisms of zinc oxide nanoparticles in the improvement of the radiosensitivity of lung cancer cells in clinically relevant megavoltage radiation energies in-vitro

Objective(s): Semiconductor zinc oxide nanoparticles (ZnO NPs) have unique properties, such as inherent selectivity and photosensitization effects under ultraviolet (UV) radiation. ZnO NPs serve as promising anticancer agents. However, UV radiation limits their penetration into the body. In most clinical settings, it is essential to use high-energy photons in the treatment of deep-seated tumors...

Dihydroartemisinin increases radiosensitivity of A549 lung cancer cells

Background: Radiotherapy is the gold standard in the treatment of lung cancer. However, the radiosensitization of cancerous cells requires further improvement. Here, we investigated the effect of dihydroartemisinin (DHA) on the radiosensitization of non-small cell lung cancer (NSCLC) cells. Methods: Cell proliferation and cell cycle assays were carried out using A549 cells exposed to DHA. The e...

Expression of Epidermal Growth Factor Receptor and the association with Demographic and Prognostic Factors in Patients with Non-small Cell Lung Cancer

Introduction: Growth, proliferation, survival, and differentiation are the prominent characteristics of cells, which are affected by cancer. Epidermal growth factor receptor (EGFR) plays a pivotal role in the effective control of these features. Given the significance of EGFR signaling pathway in non-small cell lung cancer (NSCLC), EGFR expression is influential on these cell characteristics. I...

Background and Aim: Lung cancer is the most common cause of cancer-related deaths wordwide and non-small cell lung cancer (NSCLC), especially adenocarcinoma, is the most common type of lung cancer. Most cases of adenocarcinoma will occur due to KRAS mutations or EGFR mutations and amplifications. The polymorphism rs884225 which is associated with increased EGFR expression is located in the 3'UT...

Radiosensitization effect of ZnO nanoparticles in lung cancer cells at clinically relevant megavoltage energy

Introduction: Radiation therapy is one of the major modalities that have long been used in cancer treatment. Radiotherapy is often accompanied by early and late toxicity and side effects and narrow therapeutic window. Similarity in radiation absorption properties of tumors and neighboring healthy tissues is often the reason for low specificity of radiation therapy. Development ...
